Monitoring Cell Proliferation

This lecture and lab considers different methods of tracking immune cell function by flow cytometry with a focus on monitoring proliferation using dye dilution techniques. The laboratory will address critical issues for these assays. Participants will by “hands on” experience learn how to stain and analyze cells using two different types of cell tracking dyes- CFSE and PKH26. We will also cover several instrument setup and data collection issues likely to be of interest even if your laboratory is not already doing functional assays. We will end with a discussion of different ways to analyze these data and examine a set of 4-color data correlating cytokine expression, tetramer binding and differential lymphocyte subset proliferation.
